MDP-independent activation of NF-κB by NOD2-C2. Relative NF-κB activity was measured using a luciferase-based NF-κB reporter assay.
HEK293 cells were cotransfected with NOD2-C2 (A), NOD2 or NOD2-C2/NOD2 (B) expression
constructs and dual-lucifease reporter plasmid. 10 μg/ml MDP were used to stimulate
NOD2-mediated NF-κB activation. Relative luciferase activity (compared to NOD2 (2ng)
without MDP stimulation) is plotted against NOD2 and/or NOD2-C2 plasmid amounts. Values
represent the mean ± standard deviation of triplicate cultures. Statistically significant
differences are marked by asterisk * p < 0.05; ** p < 0.01.
