Figure 3.

Notch1 intracellular domain downregulates Snail protein. (A) Hep3B cells were transfected with Myc-Notch1 intracellular domain (NICD) and/or hemagglutinin (HA)-Snail and evaluated for the expression of both proteins using immunoblot analysis. (B) Hep3B cells were transfected with Myc-NICD and/or HA-Snail and evaluated for the expression of Snail mRNA using RT-PCR. (C) Hep3B cells were transfected with HA-Snail and/or different doses of Myc-NICD, then evaluated for the expression of both proteins using immunoblot analysis. β-actin served as an internal control. (D) Hep3B cells were transfected with HA-Snail and/or different doses of Myc-NICD and evaluated for the expression of Snail mRNA using RT-PCR. β-actin served as an internal control. (E) Hep3B cells were transfected with Myc-NICD and/or treated with 300 μM H2O2 for 72 hours, then evaluated for the expression of NICD and Snail proteins using immunoblot analysis. Densitometry results of Snail analysis are shown in the right panel bar graph. (F) Hep3B cells were transfected with Myc-NICD and/or treated with 300 μM H2O2 for 72 hours, then evaluated for Snail mRNA expression using RT-PCR. β-actin served as an internal control. Bar graphs show the means ± SD of three independent experiments.
