A) Simulation of the kinetic model based on a fixed initial condition (provided in [15]) represents measurement at the single cell-resolution of the system. The response curves of serum stimulation on the different genes in the model are plotted. B) Assign a Gaussian distribution with small variances on the initial gene expression level of the untreated cells to represent the cell-cell variation in a clone. Simulation-results are computed by averaging the responses of 100 cells in a clone. C) Sampling of 100 clones under different, randomly generated initial conditions. The simulation results are shown as histograms of the expression level of the different genes, together with their ΔAIC values.
