Edited miRNAs. A) miRNAs of which at least 10% was found to be edited. Shown are the precursors in their
predicted folding structure. Red sequences represent miRNAs that are covered in our
DGE profiles, bold "a"s along numbers indicate known and newly detected editing positions.
B) Fractions of edited vs consensus miRNAs. Columns represent duplicates from each tissue,
rows indicate miRNAs and edited position on the precursor. The red intensity reflects
the fraction of edited miRNAs (edited miRNAs on this position/(edited + consensus
miRNAs)). The triplets on the right side reflect the wild-type allele of which the
middle adenosine is edited. C) qPCR data for mir-377. The first two bars describe results obtained from brain, the only tissue from which
mir-377 was cloned. The ΔCt in brain corresponds to a ~2.7 fold higher expression of the consensus
compared to the edited miRNA. Testis served as a negative input control. The specificity
of the primers is indicated by the synthetic consensus and edited miRNA, mixed in
a 1:4 ratio (cs: consensus, ed: edited). nc: primers only.
