Embryos were collected, and stained with DAPI, mAb414 to recognize nuclear pore complex and anti-gamma tubulin to recognize the centrosome. Spinning disk confocal microscopy was used to obtain Z-stack images of the prophase nucleus. The movie (QuickTime Player 7.0.3) depicts a 10.0 μm region that includes the nucleus cross-sectioned every 0.2 μm for a total of 50 cross sections. DAPI, mAb414 and anti-gamma tubulin images were merged to evaluate prophase chromosome location relative to the nuclear membrane.
