ALIX mediated rescue of HIV-1 L-domain mutant occurs independently of POSH. Endogenous levels of POSH were knocked down by RNAi in order to determine, whether ALIX can enhance virus release in the absence of POSH. HIV-1_ΔPTAP was co-transfected with a control siRNA together with either an empty plasmid or an ALIX expression plasmid (lanes 1 and 2 respectively).
