Lipid rafts are microdomains within the plasma membrane in which the lipids are more ordered than in the surrounding bilayer and they play an important role in processes such as signalling, membrane fluidity and protein trafficking. Staining methods for visualizing these lipid domains have in the past been developed almost exclusively for animal cells and tissues. Here the authors describe procedures they have developed and verified for the successful fluorescent staining of lipid domains in living leaf tissue and protoplasts.
