This is an innovative application of requencing technology to obtain full mitochondrial sequences, thus obtaining the full mitochondrial DNA sequences from 14 species, including 5 members of the Anopheles punctulatus sibling species group. Results indicate that the ancestors of the punctulatus group arrived in Papua New Guinea 20 to 40 million years ago and rapidly diverged to form the current sibling species.

Purification of Plasmodium sporozoites is an important need, for all of the reasons presented by the authors, in particular the need for clean sporozoites for human immunization trials. It seems likely that the method described will be useful for biochemical, molecular biological and in vitro studies, especially as it permits in vitro studies of P. vivax hypnozoites.

The population pharmacokinetic properties of piperaquine were described by a three-compartment disposition model in pregnant and non-pregnant women with uncomplicated malaria. The modelling approach showed no major difference in piperaquine exposure between the two groups and data presented do not warrant a dose adjustment in this vulnerable population.
