Flow cytometric analyses of stimulated CD4⁺ T cells. Purified CD4⁺ T cells were stimulated with PMA and ionomycin for 4-5 h in the presence of GolgiStop (monensin). The expression of CD300a/c was determined in the T_CM (CD45RO⁺CD27⁺) and T_EM (CD45RO⁺CD27^-) cytokine producing cells. A representative healthy donor is shown for (A) the IFN-γ, IL-4 and IL-17a, and (B) IL-2 and TNF-α producing cells. Each graph represents the percentage of CD300a/c⁺ and CD300a/c^- T_CM and T_EM cells that produce a given cytokine. Each symbol corresponds to a different donor.
