Precision (right-hand y axis scale) of the mean velocity from all cells in an image at a single time point is represented by the square root of the average variance from triplicates ranked according to mean velocity for either KM101 cells (square symbols – solid line) or MG-63 cells (round symbols – dashed line)(See Methods section). The bars in the background represent the number of triplicate sets that were included in each velocity bin (x axis) for determining average variance for KM101 cells (black bars) and MG-63 cells (white bars). The reliability of the precision estimates is reinforced by the smooth correlation of precision estimate with mean velocity across much of the range. Note how precision estimates became more "unstable" as the number of triplicate sets (bars in background) decreased at higher mean velocities.
