p35 binds to DNA fragment containing ACS like elements from c-myc origin region. A 300 bp DNA fragment containing two ACS like elements (10/11 matches) was PCR amplified using primers specific for the zone of replication from c-myc locus. PCR product was end labelled using γ-³²P ATP and used for gel shift assays. Competition reactions were performed by using different quantities (50×, 100× and 200×) of 50 bp long double stranded oligos (oligo 'b', Fig. 7B) in the reaction mixtures corresponding to lanes 3–5. Lanes 6–8 contained 65 bp double stranded DNA fragment (plasmid BlueScript KS(+) digested with HinfI followed by gel purification).
